Electroporation application development substantially affected the development of research process including controlling cell functions by using pulses of electrical fields to create pores through a cellular membrane causes celllysis and apoptosis commonly known as electroporation. Here we demonstrate the influence of the μsPEF on the Breast cancer cells (MCF-7) membrane. The purpose of this study is to evaluate the effects of various electroporation parameters on MCF-7 cells. While the cultured MCF-7 cells are placed in 9 mm-gap EP electrode chamber to allow real time observation of membrane permeability changes and cellular physiology. In order to initiate higher cell viability rate, high transfection efficiency, lower sample contamination and smaller Joule heating effect the modification of EP chamber need to be done which can be controlled by pH scale, temperature and humidity. They were treated with various pulse parameters of electroporation, including 500V/cm, 1ms; 1000V/cm, 30μs; 1500V/cm, 30μs; 2000V/cm, 30μs; and 2500V/cm, 30μs. Each parameter had two experiment settings; first setting cells were excited with single pulse and the second setting cells were excited with two pulses, one-second interval in between pulses. Our results indicate that the experimental with two pulses is more efficacious than the experimental with one pulse, and the results indicate that the optimum parameter is 2500V/cm, 30μs with two pulses with onesecond interval in between pulses.
